Production of spiculisporic acid by Talaromyces trachyspermus in fed-batch bioreactor culture

Abstract Spiculisporic acid (SA) is a fatty acid-type biosurfactant with one lactone ring and two carboxyl groups. It has been used in metal removers cosmetics, because of its low propensity to cause irritation the skin, anti-bacterial properties, high surface activity. In present study, we report an effective method for producing SA by selecting high-producing strain investigating medium components, conditions, environments culture. Among 11 kinds Talaromyces species, T. trachyspermus NBRC 32238 showed highest production crystalline substance, which was determined be using NMR. The able produce under acidic conditions from hexoses, pentoses, disaccharides, glucose sucrose serving as most appropriate substrates. Investigation nitrogen sources trace ions revealed meat extract FeCl 3 components that promoted production. Upon comparing types cultures baffle flask or aeration bioreactor, found slightly higher than reactor. bioreactor culture, substrate production, compared glucose, sucrose, lag time until start shortened. Finally, fed-batch culture resulted 60 g/L SA, total yield 0.22 g SA/g productivity 6.6 g/L/day.
